The ability todetect such targets, especially HABspecies and toxins, in real or nearrealtime is of immediate utility toresearchers studying HAB/toxindynamics and to coastal managerscharged with mitigating thesocioeconomic impacts of blooms. The Environmental SampleProcessor (ESP;www.mbari.org/microbial/ESP) wasdeveloped for the autonomous, subsurfaceapplication of moleculardiagnostic tests and during initialdeployments has successfully runDNA probe arrays for several HABspecies. To address the need forconcurrent detection of algal toxins, which are known to vary widely inconcentration depending on algalphysiological status, we recentlyfielded a toxin extraction protocolcoupled with immuno-based toxinarrays during deployments of asecond generation ESP. Throughapplication of species and toxinarrays, known toxic Pseudonitzschiaspp. and domoic acid (DA)were detected concurrently onboardthe ESP in Monterey Bay, CA, USA,representing the first remote, integrated assessment of algal cellabundance and toxin concentrationin coastal waters. Refinedcalibration of the DA assay will yieldmore accurate estimates of abloom’s toxicity and potentialimpacts.O.16-02The detection of toxic algae by anew developed rRNA biosensor(EU-Project ALGADEC)Session: O.16 - Monitoring 1Presentation time: 12:00 - 12:20S Diercks, K Metfies, LK MedlinAlfred Wegener Institute, BREMERHAVEN,GermanyThe EU-project ALGADEC aims todevelop a hand-held biosensor forin situ analysis of toxic algae whichcan serve as an early warningsystem to prevent toxication ofanimals and consumers.Identification of toxic algae is basedon molecular probes that specificallytarget its rRNA. Taxon- specificprobes were developed for HABalgae that occur in three differentcoastal areas in Europe. Asandwich-hybridization assay isused to detect the rRNA.
