Inorder to investigate itspharmacology, a requirement forthe producti<strong>on</strong> of gymnodimine isthe guarantee of a reliable andenhanced supply of the compound.We have c<strong>on</strong>structed aphotoreactor (15 L capacity)allowing improved light access (190μmol/m 2 /s) operated in a 12hlight:12h dark cycle, with gentlestirring and aerati<strong>on</strong> of culture. Theunit also has an inlet for carb<strong>on</strong>dioxide injecti<strong>on</strong>, and capability ofpH and turbidometricmeasurements. We have operatedthe reactor <strong>on</strong> a semi-automaticbasis with pH poised at 8.5. Growthkinetics and gymnodimineproducti<strong>on</strong> by K. selliformis (growthyield maximum, 46510 cells/ml;doubling time, 8.6 days;gymnodimine, 0.5 μg/ml) weresubstantially improved compared tostandard (12 L) batch culture(growth yield max. 41,510 cells/ml;doubling time, 17.3 days;gymnodimine, 0.16 μg/ml) with nopH amendment. As a c<strong>on</strong>sequencea fully automated culture systemhas been developed. Although theoriginal Texas brown tide bloompersisted 1990-1997, there is <strong>on</strong>lylimited informati<strong>on</strong> <strong>on</strong> the growth ornutriti<strong>on</strong>al characteristics of thecausative organism. Weinvestigated the effect that organicnitrogen availability and reducedphotosynthetically availableradiati<strong>on</strong> (PAR) have <strong>on</strong> the growthof TBT. Such parameters seemcritical for blooms of themorphologically and geneticallysimilar east coast brown tidespecies Aureococcusanophagefferens. We tested theability of TBT to utilize DIN (NO 2 - orNH 4 + ) and DON (urea or glutamicacid) as the sole source of nitrogenunder 8 light intensities ranging fromlimiting to saturating irradiancesusing a light gradient table.
