Previousstudies <strong>on</strong> the species dynamicssuggest that bloom initiati<strong>on</strong>occurred in Lisb<strong>on</strong> Bay in aretenti<strong>on</strong> area related to the CapeRoca upwelling plume. During 2005,in this Bay, G. catenatum wasobserved up by the m<strong>on</strong>itoringprogramme from mid-July. InAugust/September, a survey carriedout <strong>on</strong> the area showed that G.catenatum was distributed (>500cells/L) al<strong>on</strong>g a str<strong>on</strong>g thermal fr<strong>on</strong>t,resulting from the interacti<strong>on</strong> of thecolder upwelled waters of CapeRoca plume and oceanic waters.Satellite-derived sea level anomalymaps revealed that during thecruise, and for at least three weeksbefore, the off-shelf surfacecirculati<strong>on</strong> was dominated by thepresence of two counter rotatingmesoscale eddies, resp<strong>on</strong>sible for astr<strong>on</strong>g northeastward flow of warmoceanic waters into the survey area.During autumn, the m<strong>on</strong>itoringprogramme allowed the following ofa northward shift of the populati<strong>on</strong>related to a poleward surface flow.On 29 November, G. catenatumreached the maximumc<strong>on</strong>centrati<strong>on</strong> (43x10 3 cells/L) <strong>on</strong>the Aveiro coast and lasted innorthern Portuguese waters untilJanuary 2006.PO.05-06First evidence of DTX2 inFrance: detecti<strong>on</strong> by LC-ESI-MS2during 2004-2005 south Brittanyphytoplankt<strong>on</strong> bloomsSessi<strong>on</strong>: PO.05 - Toxin analysisFlorence M<strong>on</strong>deguer, Elizabeth Nézan,Dominique Le Gal, Claire MarcaillouIFREMER, NANTES, FranceOur study relates to phytoplankt<strong>on</strong>samples collected during <strong>on</strong>e year<strong>on</strong> the southern Brittany coast(05/10/04-07/18/05). For eachsample, we worked at the sametime <strong>on</strong> tax<strong>on</strong>omic and toxin profiles(lipophilic phycotoxins). To ensurethat all samples were treatedidentically for optimumreproducibility, all phytoplankt<strong>on</strong>extracts were quickly purified <strong>on</strong>Solid Phase Extracti<strong>on</strong> (SPE), witha robotic stati<strong>on</strong> (ASPECXli,Gils<strong>on</strong>). The simultaneousidentificati<strong>on</strong> and quantificati<strong>on</strong> oftoxin profiles were achieved byusing a liquid chromatography–tandem mass spectrometry (LC–MS/MS) with electrospray i<strong>on</strong>isati<strong>on</strong>(+) and m<strong>on</strong>itoring of daughter i<strong>on</strong>sin multiple reacti<strong>on</strong> modes. Theresults obtained in the tax<strong>on</strong>omicanalysis showed that Dinophysisacuminata was the predominantspecies (75%). The toxin analysis ofthe samples by LC-ESI-MS/MSshows that okadaic acid (AO) is theprincipal lipophilic toxin. Howeve,r02/11/04 a m<strong>on</strong>ospecific samplec<strong>on</strong>taining 75% of Dinophysisacuta, was an excepti<strong>on</strong> to theuniformity of the profiles observedduring this year. The LC-MS2analysis of this plankt<strong>on</strong> net towshows the rare presence of DTX2(dinophysistoxin-2). For this sample242
