CARLESDE LA RÀPITA, Spain2 Instituto Español de Oceanografía, VIGO,SpainIn July 2005, a bloom of thedinoflagellate Protoceratiumreticulatum at cell concentrationlevels of 1600 cells/L was detectedin Alfacs Bay, in conjunction withthe presence of Dinophysissacculus.Positive DSP mouse bioassaymeasurements indicated thepresence of diarrhetic toxins inmussels. Analytical procedureswere optimised and applied tophytoplankton and to musselextracts. Mussel samples were firstextracted in methanol/H2O (80:20),further percolated through anoctadecylsilica cartridge, derivatizedusing the dienophile fluorescencereagent, DMEQ-TAD, andsubsequently analysed by liquidchromatography coupled tofluorescence detection (LC-FD).Concentration levels of yessotoxinin mussels about 0.3 to 1.0 µg/kgwere detected during this event.Phytoplankton samples wereextracted by sonication withmethanol, followed by a clean-upstep. Okadaic acid concentrationlevels in these samples were alsostudied to confirm the toxic effectsrecorded along theProtoceratium/Dinophysis bloom.Evaluation of cell-based toxicity offield samples by estimation of cellviability, IC 50 and morphologicaleffects was studied and comparedto the analytical measurements.Protoceratium reticulatum waspreviously detected inphytoplankton samples from theEbro Delta embayments in summer2001, and concentration levels ofyessotoxin and homoyessotoxinwere then confirmed in musselsamples.PO.13-45The influence of Pseudonitzschiaaustralis blooms inshellfish domoic acidaccumulation on the Andalusiancoast (southern Spain)Session: PO.13 - Regional eventsLuz Mamán, D Jaén, R Fernández, MAOcaña, l Fernández, I MarquezLaboratorio Control Recursos Pesqueros,HUELVA, SpainDiatoms of the genus Pseudonitzschiahave been detected at theAndalusian coast since themonitoring program began in 1996.Some species from this genus areharmful because of the productionof the toxin domoic acid. During2006, samples from the monitoringprogram were analysed with LMand EM, allowing identification ofPseudo-nitzschia australis as themost abundant Pseudo-nitzschiaspecies. This event resulted inmarked toxin accumulation inseveral bivalve molluscs from theMediterranean coast.231
