Pyrodinium bloomsare often unpredictable andsometimes occur at very lowdensities. One of the problems inthe m<strong>on</strong>itoring and management ofthe Pyrodinium red tides is the earlydetecti<strong>on</strong> of the species. This studyinvolves the development of a DNAprobe from highly c<strong>on</strong>served smallsubunit rDNA regi<strong>on</strong>s for fast andaccurate identificati<strong>on</strong> of the alga.Seawater samples c<strong>on</strong>tainingPyrodinium taken from four offshorelocati<strong>on</strong>s were isolated and culturedin f/2 medium. After DNA extracti<strong>on</strong>,PCR amplificati<strong>on</strong> was carried outtargeting the rDNA of the algae. Theresults of eight restricti<strong>on</strong> enzymedigesti<strong>on</strong>s indicated that all thecultures were similar. Sequences ofother Pyrodinium isolates were alsoobtained (Accessi<strong>on</strong> number:DQ500119 - 23). The size of thesequences ranged from between1580 to 1544 bp.
