Apopitosisin düzenlenmesi önemli miktarda effektör ve regülatör bileşken içerir.<br />
Programlanmış hücre ölümünün önemine rağmen, apopitosisin mekanizmaları ve<br />
apopitosis ile ilgili birçok moleküler etkileşim henüz tam olarak açıklığa kavuşmamıştır.<br />
Çalışmamızda Drosophila melanogaster da proapopitotik genler olan grim ve hid<br />
genler aracılığıyla düzenlenen apopitosisde Nitrik Oksit’ in (NO) rolü incelenmiştir.<br />
Normal Drosophila metamorfozunun larval imeginal diskte yüksek proliferasyon hızı<br />
ve apopitosisin bulunmamasıyla karakterize olduğuna inanılmaktadır. Bundan dolayı<br />
, olgun bir sineğin gelişimi imaginal hücrelerdeki değişimin sonucuna bağımlıdır.<br />
Dolayısıyla Drosophila gelişiminin larval periodu esnasında imaginal disk hücrelerinin<br />
apopitotik hücre ölümü strese maruz kalmayla ilişkilidir. Çalışmamızda NO nun aşırı<br />
üretimi esnasında larvaların transgenik türlerinin NO sağlayıcısı olarak proapopitotik<br />
grim and hid genelerinin expresyonunu artırdığı ve yeni baştan (de novo) imaginal disc<br />
te apopitosisin indüklendiği gösterilmiştir. Buna karşılık, düşük NO üretimi esnasında<br />
transgenik larva türleri NO inihbitörü gibi etki gösterdiğinde grim ve hid genlerin<br />
ekspresyonun azaltılmış, ve apopitotik süreç baskılanmıştır. Drosophila melanogaster<br />
de apopitotik sürecin düşük miktardaki NO ile zayıflatılır iken yüksek NO hücre içi<br />
konsantrasyonlarının güçlü bir apopitosis indükleyicisi olduğu öne sürülebilir.<br />

Apoptosis regulation involves large number of regulatory and effector components.<br />
Despite the importance of programmed cell death, many molecular interactions<br />
and mechanisms of apoptosis are not yet fully clarified. In our study we examined<br />
participation of nitric oxide (NO) in regulation of apoptosis mediated by proapoptotical<br />
grim and hid genes in Drosophila melanogaster. It is believed that normal Drosophila<br />
metamorphosis is characterized by high proliferation rate and absence of apoptosis in<br />
larval imaginal discs. It is due to the fact that development of an adult fly is the result of<br />
differentiation of imaginal cells. Therefore, apoptotic cell death of imaginal discs cells<br />
during larval period of Drosophila development is associated with stress exposure. In<br />
our study we demonstrated that during the period of NO overproduction in larvae of<br />
transgenic lines acting as NO donors expression of proapoptotic grim and hid genes<br />
increases and in imaginal discs apoptosis is induced de novo. Whereas in larvae of<br />
transgenic lines acting as NO inhibitors, during the period of low NO production<br />
expression of grim and hid genes is reduced, and apoptotic processes are suppressed.<br />
We assume that high cellular concentration of NO act as a potent apoptosis inducer,<br />
while low doses of NO can lead to attenuation of apoptotic processes in Drosophila<br />
melanogaster.<br />
