POLYMORPHISM IN TURKISH POPULATION<br />

Araşidonik asit türevi olan Epoksieikosatrienoik asitler (EET’ler) güçlü<br />
vazodilatördürler ve anti-hipertansif, anti-platelet, anti-inflamatuar etkileri vardır.<br />
Soluble epoksit hidrolaz(sEH) EETleri de içeren yağ asidi türevi epoksitleri<br />
metabolize eden en önemli enzimdir. EETler sEH tarafından yıkılarak deaktive<br />
edilirler. Son zamanlarda, EET’lerin lipit metabolizmasındaki önemli rolü nedeniyle<br />
sEH hipertansiyon ve kalp krizi gibi hastalıkların tedavisinde bir hedef molekül haline<br />
gelmiştir. Hipertansiyonun tedavisi için yeni ilaçlar elde edilmesi amacıyla sEH<br />
inhibitörleri geliştirilmiştir. sEH enzimini kodlayan gende (EPHX2) birkaç genetik<br />
polimorfizm belirlenmiştir. Proteinde Arg287Gln aminoasit değişimine neden olan<br />
G860A polimorfizminin enzim aktivitesinde azalmaya neden olduğu bildirilmiştir.<br />
Bazı çalışmalar EPHX2’deki genetik farklılıkların kardiyovasküler hastalıklarla<br />
ilişkisine dikkat çekmişlerdir. Bu çalışmada hipertansiyon ve kardiyovasküler hastalık<br />
sıklığının oldukça yüksek olduğu Türk popülasyonunda EPHX2 G860A polimorfizm<br />
sıklığını belirledik. Çalışılan popülasyonda yabanıl tip (GG), heterozigot (GA) ve<br />
homozigot polimorfik (AA) genotip sıklıkları sırasıyla %79 (n =237), %20 (n=61) ve<br />
%1 (n=2) olarak belirlendi. EPHX2 G860A polymorfizmi genotip sıklıkları Hardy-<br />
Weinberg dengesindeydi. Genotip sıklıklarımızın Çin popülasyonundan farklı iken,<br />
Danimarka, İsveç ve Afrikalı-Amerikalı popülasyonları ile benzer olduğu gözlendi.<br />
Epoxyeicosatrienoic acids (EET’s) derived from arachidonic acid are potent<br />
vasodilators that have antihypertensive, antiplatelet and antiinflamatory effects.<br />
Soluble epoxide hydrolase (sEH) is the primary enzyme that metabolises fatty acid<br />
derived epoxides including EETs. The breakdown of EETs by sEH is in general<br />
belived to be a deactivation process. Due to it’s important role in the lipid metabolism<br />
of EETs the sEH has recently evolved as a target for the treatment of hypertension,<br />
inflamatory deseases and stroke. Inhibitors of the sEH have been developed aiming<br />
in particular at new therapeutics for the treatment of hypertension. Severeal genetic<br />
polymorphism have been reported for the gene (EPHX2) encoding the sEH enzyme.<br />
The G860A substitation resulting in an amino acid change at position 287 of the protein<br />
(Arg287Gln) causes a significant decrease in the activity. Several studies have addressed<br />
the association between different EPHX2 genetic variations and cardiovascular<br />
diseases. In this study we determined the EPHX2 G860A polymorphism frequency<br />
in Turkish population having very high frequency of hypertension and cardiovascular<br />
diseases. The frequencies of the wild type (GG), heterozygous (GA) and homozygous<br />
polymorphic (AA) genotypes in the study population were 79% (n =237), 20%<br />
(n=61) and 1% (n=2) respectively. The genotype frequencies for EPHX2 G860A<br />
polymorphisms were in Hardy-Weinberg equilibrium. Our genotype frequencies were<br />
significantly different from those reported for Chinese population and very similar to<br />
those reported for Danish, Swedes and African-American populations.<br />
