Süperoksit dismutaz–1 (SOD–1) geni, 21. kromozomda haritalanmakta ve enzimin<br />
aktivitesi Down sendrom (DS) ’lu bireylerin hücrelerinde yaklaşık %50 oranında<br />
artmaktadır. SOD’dan sonraki basamaklarda yer alan antioksidan enzimlerin bu artışı<br />
kompanse edemediği DS’lu kişilerde, oksidatif stresin arttığı düşünülmektedir. Biz<br />
de DS’lu hasta grubunda, sağlıklı kontrol grubuna göre oksidatif stres düzeylerini<br />
değerlendirmeyi amaçladık.<br />
Bu çalışmaya, hasta grubu olarak 37 (13 kadın/24 erkek) DS’lu birey ile kontrol<br />
grubu olarak 42 (13 kadın/29 erkek) sağlıklı birey dahil edildi. DS’lu olguların yaş<br />
ortalaması 4,00 ± 2,93 iken, kontrol grubunun yaş ortalaması ise 5,07 ± 3,31 yıl olarak<br />
belirlendi. Çalışmada, hasta ve kontrol grubunda Total Antioksidan Statü-TAS (Erel<br />
yöntemi, Rel Assay), Total Oksidan Statü-TOS (Erel yöntemi, Rel Assay), SOD<br />
(19160 SOD determination kit, Sigma-Aldrich) ve Glutatyon Peroksidaz -3- GPx-3<br />
(ELISA, Adipogen) parametrelerinin serum düzeylerine bakıldı.<br />
Kontrol grubunun ortalama TAS (1,66±0,24 Trolox Eq/L) ,TOS (10,72±5,04 umol<br />
H2O2 Eq/L), GPx-3 (9,54±4,99 ng /mL) ve SOD düzeyleri (3,18±1,23 U/ml) ile hasta<br />
grubunun ortalama TAS (1,59±0,26 Trolox Eq/L) ,TOS (11,81±9,55 umol H2O2<br />
Eq/L), GPx-3 (9,89±4,35 ng /mL) ve SOD (3,26±1,02 U/ml) düzeyleri arasında<br />
istatiksel olarak anlamlı bir fark bulunamadı (p > 0,05).<br />
Sonuç olarak, biz DS’lu bireylerde SOD, GPx-3, TAS ve TOS düzeylerinin sağlıklı<br />
kontrol grubuna ait değerlerde farklı bulmamamıza rağmen, bu parametrelerin ölçümü;<br />
SOD-1 intrasellüler bir enzim olduğu için izole hücrelerin sitoplazmik örneklerinde<br />
ya da oksidatif stres yaşla arttığı için daha ileri yaş gruplarında yapılırsa daha anlamlı<br />
sonuçların elde edileceği kanısına vardık. Bu parametrelerin, DS’da oksidatif stres<br />
ile ilişkilendirilen patolojilerin değerlendirilmesi, takip ve tedavisinde faydalı olacağı<br />
inancındayız.<br />
Superoxide dismutase–1 (SOD–1) gene is mapped on the 21st chromosome and<br />
activity of the enzyme increases approximately by 50% in the cells of the individuals<br />
with Down syndrome (DS). It is thought that oxidative stress increases in those people<br />
with DS, antioxidant enzymes of whom cannot compensate for this increase at those<br />
steps after SOD. We aimed at evaluating the oxidative stress levels of the patient<br />
group with DS compared to the healthy control group.<br />
37 individuals (13 women/24 men) have been included as the patient group and 42<br />
healthy individuals (13 women/29 men) have been included as the control group to<br />
the study. Average age of the adults with DS has been determined as 4.00 ± 2.93;<br />
whereas, the average age of the control group has been determined as 5.07 ± 3.31<br />
years. In the study, serum levls of the Total Antioxidant Status – TAS (Erel method,<br />
Rel Assay), Total Oxidant Status – TOS (Erel method, Rel Assay), SOD (19160 SOD<br />
determination kit, Sigma-Aldrich) and GPX-3 (ELISA kit, Adipogen) parameters<br />
have been examined.<br />
A statistically meaningful difference between the TAS (1,66±0,24 Trolox Eq/L) ,TOS<br />
(10,72±5,04 umol H2O2 Eq/L), GPx-3 (9,54±4,99 ng /mL) and SOD levels (3,18±1,23<br />
U/ml) of the control group and TAS (1,59±0,26 Trolox Eq/L) ,TOS (11,81±9,55 umol<br />
H2O2 Eq/L), GPx-3 (9,89±4,35 ng /mL) and SOD levels (3,26±1,02 U/ml) of the<br />
patient group has not been detected (p > 0,05).<br />
Consequently, although we didn’t find SOD, GPx-3, TAS and TOS levels of the<br />
individuals with DS different than the values of the control group, we have concluded<br />
that if measurement of these parameters is done on the cytoplasmic samples of the<br />
isolated cells because SOD-1 is intracellular or on older age groups because oxidative<br />
stress increases with age, more meaningful results can be achieved. We believe<br />
that these parameters shall be useful in evaluation, monitoring and treatment of the<br />
pathologies associated with oxidative stress of DS.<br />
