mtDNA Real Time PCR analizi biyolojik delillerin değerlendirilmesinde çok önemli<br />
bir araç olarak ortaya çıkmaktadır. Özellikle az miktardaki örneklerde başvurulması<br />
gereken ileri derecede bir incelemedir. Günümüzde, çeşitli mtDNA polimorfik bölgeleri<br />
Adli Bilimler uygulamalarına kazandırılmaktadır. Bu durumda mtDNA’nın son derece<br />
yüksek bir dışlama gücüne ulaşacağı kesindir. Ancak, mtDNA genotipleme analizi<br />
yeni bir sistem olduğundan, yeni mutasyonların adlandırılmasından, heteroplazmi<br />
olayının izlenmesine kadar her noktanın dikkatle uygulanması gerekmektedir.<br />
Avrupa popülasyonu düşünüldüğünde kişilerarası ortalama 8 nükleotidte farklılık<br />
görülmektedir. Bu 8 farklılıktan biri, 7025. bazda meydana gelen polimorfizmdir.<br />
Bu bölgenin sağlıklı 10 gönüllüden aldığımız serum numunelerinde polimorfik olup<br />
olmadığının oldukça hızlı ve güvenilir sonuç veren ve floresans işaretli primerlerin<br />
kullanılması prensibi esası ile çalışan Real Time PCR cihazı ile analizine dayanarak<br />
bir ön tarama çalışması yapmayı hedefledik.<br />
Her ne kadar çalışmamızda denek grubu 10 kişiden oluşmuş olsa da, başarılı bir<br />
genotipleme yapılabilmiş ve sonuçlar literatür bilgisi ile karşılaştırılmıştır. Buna göre,<br />
GT kontrol SNP, ilk olarak ve çoğunlukla bilinmeyen mtDNA örneklerinin kalitesini<br />
araştırmak için kullanılmaktadır. Bu SNP’nin kullanımı ile, 10 mtDNA örneğinden<br />
8’ini başarı ile genotipledik. 2 mtDNA örneğinden düşük floresans sinyal elde ettik. 10<br />
numaralı örnek, OD ölçümüne göre çok düşük miktarda mtDNA içermekteydi. Elde<br />
ettiğimiz tüm Real-Time analiz sonuçlarına göre, farklı genotipler Amplifluor Real-<br />
Time metodu kullanılarak belirlenebilmiştir. Sonuçlar, konvansiyonel Amplifluor<br />
end-point metodu ile konfirme edilmiştir.<br />
Dünya çapında birçok laboratuarda idantifikasyon amacı ile sıklıkla başvurulan bu<br />
yöntemle ilgili olarak ülkemizde faaliyette bulunan kriminal laboratuvarlarda hala<br />
ciddi bir çalışma yoktur. Bu nedenle çalışmamızda yöntemin standardizasyonunun<br />
sağlanması ve bu mutasyonla ilgili bir ön bilgi edinilmesi amaçlanmıştır.<br />
It is not always possible to obtain enough nuclear DNA sample for the analysis of<br />
the biological samples in the purpose of forensic evaluations. The polymorphic DNA<br />
found in the mitochondria matrix exist at least a hundred copies and it makes easy<br />
to obtain results. Where it is not possible to work with nuclear DNA, mtDNA gives<br />
successfull results. The aim of this study was to have a preliminary information for<br />
the genotyping of the mtDNA and to obtain data about the mutation in 10 healthy<br />
volunteers.<br />
The development of a one step, PCR-based, universal detection method, the<br />
Amplifluor® SNPs Genotyping System, simplifies high throughput analysis for the<br />
genotyping of SNPs. The Amplifluor® SNPs Genotyping System is a highly sensitive,<br />
homogeneous fluorescent assay. It can be adapted to virtually any sample that can be<br />
analyzed by allele-specific PCR.<br />
The 7025th SNP site of the mtDNA was genotyped with Real-Time PCR instrument<br />
and the haplotypes were evaluated compared to standart end-point PCR results for<br />
understanding the applicability of Real-Time PCR in genotyping mtDNA for forensic<br />
purposes. Blood samples were collected from 10 unrelated individuals and mtDNA<br />
was extracted with a commercially available kit. The target regions were amplified by<br />
means of Real-Time PCR instrument and then the products were genotyped by using<br />
an appropriate labelling for the universal Amplifluor fluoresence method.<br />
Even if our study group had consisted of 10 volunteers, we could have a successfull<br />
genotyping for 8 samples and compare the results with the current literature.<br />
