the DNA adducts resulting from tobacco carcinogen exposure have been characterized<br />

in tissues from smokers. Other studies demonstrate convincingly that some of these<br />

DNA adducts cause mutations during DNA replication. The observed mutations are<br />

frequently similar to those seen in oncogenes <strong>and</strong> tumor suppressor genes isolated<br />

from human lung cancers. Mutations in these genes lead to cancer by disrupting<br />

normal cellular signaling processes that control growth. The studies summarized here<br />

therefore demonstrate how cigarette smoke carcinogens—through human uptake,<br />

metabolic activation, <strong>and</strong> induction of mutations—firmly link nicotine addiction to<br />

cancer in smokers.<br />
