Prerequisites/ Level Advanced Course Description The following analytic-diagnostic methods are planned: 1. Identification of oncogenic mutations in the signaling molecule KRAS � Preparation of genomic DNA from cell lines and/or paraffin-embedded and formaldehyd-fixed tumour specimen � Preparation of RNA from cell lines and/or paraffin-embedded and formaldehydfixed tumor specimen � Generation of cDNA from RNA samples � Real-Time PCR experiments to identify and characterize mutations in the KRAS oncogene 2. Pyrosequencing experiments to identify and characterize genomic mutations in the KRAS oncogene 3. Immunohistochemistry of paraffin-embedded human cancer specimen Teaching Concept Each day the course will be divided into two parts. In the first (theoretical) part the students will get detailed background information regarding the cells (origin/biology) and materials (chemical composition, applicability) employed in the course. In addition, we will discuss the advantages and shortcomings/pitfalls as well as the clinical relevance of the molecular diagnostic techniques including data interpretation and validation. In order to guarantee a precise and smooth workflow in the laboratory the students will also receive first technical instructions on the upcoming experiments.
