Universidad Complutense,28040-Madrid Tel.: 91-3941454 Fax: 91-3941691 e-mail: jara@eucmax.sim.ucm.esResumenNumerosos estudios realizados en las dos últimasdécadas han <strong>de</strong>mostrado la existencia en el organismoanimal <strong>de</strong> un sistema cannabinoi<strong>de</strong> endógeno,constituido por unos ligandos, los endocannabinoi<strong>de</strong>s.Se han <strong>de</strong>scrito dos tipos <strong>de</strong> receptores paracannabinoi<strong>de</strong>s: los <strong>de</strong>nominados CB 1 , localizados preferentementeen cerebro y los CB 2 que están localizadosen el sistema inmune. Los dos endocannabinoi<strong>de</strong>s<strong>de</strong> los que más datos se dispone son laaraquidoniletanolamida o anandamida y el 2-araquidonilglicerol,habiéndose postulado su posible actuacióncomo neurotransmisores o neuromoduladores.La distribución cerebral <strong>de</strong> los endocannabinoi<strong>de</strong>sy <strong>de</strong> los receptores CB 1 ha permitido conocer las funcionesfisiológicas en las que está involucrado estesistema. Participa, a través <strong>de</strong> modular la actividad <strong>de</strong>los neurotransmisores, en la regulación <strong>de</strong>l comportamientomotor y <strong>de</strong> la secreción <strong>de</strong> hormonas a<strong>de</strong>nohipofisarias,interacciona con la dopamina y con elGABA, mientras que, en el caso <strong>de</strong> la memoria y elaprendizaje, lo hace con el GABA y el glutamato. Ladopamina y los péptidos opioi<strong>de</strong>s podrían estar implicadosen la participación <strong>de</strong> los endocannabinoi<strong>de</strong>sen el sistema <strong>de</strong> recompensa y en el control <strong>de</strong> lanocicepción. Por otro lado, la síntesis <strong>de</strong> anandamida,en condiciones <strong>de</strong> isquemia, podría jugar un papelprotector en las regiones cerebrales afectadas. Se havisto que este compuesto inhibe la captación mitocondrial<strong>de</strong> calcio y la liberación <strong>de</strong> glutamato, efectosámbos que contribuyen a la citotoxicidad cerebral.Palabras clave: Endocannabinoi<strong>de</strong>s, anandamida,receptores CB 1 y CB 2 , transducción <strong>de</strong> señales,neurotransmisores, cannabis.SummaryNumerous studies carried out in the last two <strong>de</strong>ca<strong>de</strong>shave shown the existence in the animal organismof an endogenous cannabinoi<strong>de</strong> system comprisingcertain ligands, the endocannabinoi<strong>de</strong>s. Two types ofreceptors for cannabinoi<strong>de</strong>s have been <strong>de</strong>scribed,those known as CB1, located predominantly in thebrain, and the CB2 located in the immune system.The two endocannabinoi<strong>de</strong>s on which there is moreavailable data are the arachidonylethanolami<strong>de</strong> oranandami<strong>de</strong> and the 2-arachidonylglycerol, their possibleaction as neurotransmitters or neuromodulatorshaving been postulated.The cerebral distribution of the endocannabinoi<strong>de</strong>sand the CB1 receptors has led to knowledge of thephysiological functions that involve this system. It participatesby modulating the action of the neurotransmitters,in regulating motor behaviour and the secretionof a<strong>de</strong>nohypophysary hormones, interacts withdopamine and the GABA and, in the case of memoryand learning, it interacts with the GABA and the glutamate.The dopamine and the opioid pepti<strong>de</strong>s maybe implicated in the participation of the endocannabinoi<strong>de</strong>sin the drug reward system and in the controlof nociception. At the same time, the synthesis ofanandamine in ischemia may play a protective role inthe cerebral regions affected. It has been seen thatthis compound inhibits the mitochondrial uptake ofcalcium and the liberation of glutamate, both effectsthat contribute to cerebral cytoxicity.Key words: cannabis, endocannabinoids, anandami<strong>de</strong>,receptor, neurotransmitters, neuromodulators.VOL. 12.
