Characterization <strong>of</strong> variability <strong>in</strong> Fusarium udum : We also <strong>in</strong>itiated work on characterization <strong>of</strong> variability <strong>in</strong> F.<br />

udum. Eleven isolates <strong>of</strong> F. udum from n<strong>in</strong>e locations <strong>in</strong> six states [Akola, Badnapur (Maharashtra); Bangalore,<br />

Gulbarga (Karnataka); Khargone (Madhya Pardesh), Muradnagar (Meerut), Varanasi (UP), Warangal and<br />

<strong>ICRISAT</strong>-Patancheru (Andhra Pradesh) all <strong>in</strong> India] were collected and s<strong>in</strong>gle spore cultures were obta<strong>in</strong>ed us<strong>in</strong>g<br />

standard mycological techniques. All <strong>the</strong> s<strong>in</strong>gle spore isolates once aga<strong>in</strong> were passed through common wilt<br />

susceptible cultivar ICP 2376 by follow<strong>in</strong>g root dip <strong>in</strong>oculation technique, and aggressive isolates were stored at 4 0<br />

C <strong>in</strong> <strong>the</strong> laboratory. S<strong>in</strong>ce <strong>the</strong> collection <strong>of</strong> <strong>the</strong> isolates did not represent <strong>the</strong> entire country, we cont<strong>in</strong>ued to collect<br />

<strong>the</strong> isolates from pigeonpea grow<strong>in</strong>g areas. Dur<strong>in</strong>g <strong>the</strong> current crop season, wilt <strong>in</strong>fected samples were collected<br />

from 9 locations (Yenagandla, Kulcharam, Jogipet, Andole, and Shivampet <strong>in</strong> Andhra Pradesh; Aurad <strong>in</strong> Karnataka;<br />

and Dhuki, Osmanabad, and Parbhani <strong>in</strong> Maharashtra, India). Isolations were made and F. udum was obta<strong>in</strong>ed from<br />

all <strong>the</strong> locations. All <strong>the</strong> cultures were purified and s<strong>in</strong>gle spore isolates were obta<strong>in</strong>ed follow<strong>in</strong>g standard<br />

mycological techniques. All <strong>the</strong>se isolates were tested for virulence us<strong>in</strong>g a common susceptible cultivar ICP 2376<br />

follow<strong>in</strong>g root dip technique. All <strong>the</strong> virulent cultures were stored at 4 0 C <strong>in</strong> <strong>the</strong> laboratory.<br />

Milestone: Mapp<strong>in</strong>g <strong>of</strong> one <strong>in</strong>tra-specific RIL population for dry root resistance conduced (PMG/SP) 2008<br />

Mapp<strong>in</strong>g <strong>of</strong> RILs populations to DRR resistance under controlled environment: One hundred and twenty-six<br />

RILs and parents (JG 62 and ICCV 2) were evaluated for DRR resistance us<strong>in</strong>g paper towel technique under<br />

controlled environment conditions. Each entry was replicated thrice. Of <strong>the</strong> 126 RILs evaluated, ICCX 930111- 57-<br />

1-1-1-1SP-1-1-1-BP-1BP showed a resistant reaction (3 rat<strong>in</strong>g on 1 - 9 scale), while 15 entries were moderately<br />

resistant (3.1 to 5 rat<strong>in</strong>g). Both parents showed a susceptible reaction.<br />

Activity 6A.3.1: Advanced generation <strong>in</strong>ter-specific derivatives with resistance to Helicoverpa and BGM<br />

generated us<strong>in</strong>g wild Cicer from different gene pools<br />

Milestone: Strategies to cross wild Cicer between primary, secondary and tertiary gene pools developed (NM) 2008<br />

In collaboration with Wash<strong>in</strong>gton State University, Pullman, USA cross<strong>in</strong>g program was <strong>in</strong>itiated to cross perennial<br />

Cicer with annual as well as cultivated Cicer. Pollen germ<strong>in</strong>ation was observed between <strong>in</strong>tra-perennial Cicer<br />

species. It is possible to have <strong>success</strong>ful poll<strong>in</strong>ation and fertilization between annual species C. reticulatum and<br />

perennial species C. oxyodon and C nurstichacum, as well as between cultivated chickpea and C. oxyodon and C.<br />

nursticum, but mature seeds were not observed. S<strong>in</strong>ce perennial Cicer does not set flowers at <strong>ICRISAT</strong> Patancheru,<br />

techniques were developed to preserve pollen <strong>of</strong> perennial Cicer for use at <strong>ICRISAT</strong> at a later date. Germ<strong>in</strong>ation<br />

medium for annual and perennial Cicer pollen has also been developed.<br />

Milestone: Fifteen stable <strong>in</strong>ter-specific derivatives us<strong>in</strong>g resistant wild Cicer from secondary gene pool generated<br />

and screened for Helicoverpa, AB, BGM, and good agronomic characters under field conditions<br />

(NM/HCS/SP/PMG) 2010<br />

Cicer accessions ICC 17159, IG 73074, IG 72937, IG 72933, and IG 72934, which have shown resistance to<br />

Botrytis grey mold were used <strong>in</strong> <strong>the</strong> cross<strong>in</strong>g program. Seed set varied depend<strong>in</strong>g upon <strong>the</strong> pollen parent used <strong>in</strong> <strong>the</strong><br />

cross<strong>in</strong>g program. <strong>The</strong> F 2 progenies from <strong>the</strong> crosses <strong>in</strong>volv<strong>in</strong>g wild Cicer species IG 73074 and ICC 17159 are<br />

listed <strong>in</strong> Table 3. Chi square analysis <strong>of</strong> <strong>the</strong> progenies showed that <strong>the</strong> ratio <strong>of</strong> resistant to susceptible plants was 1:<br />

3, and hence BGM resistance <strong>in</strong>trogressed from wild Cicer was monogenic and recessive.<br />
