Milestone: 50 transgenic events <strong>of</strong> pigeonpea with maize psy1 gene developed and screened for high ß-carotene<br />

production <strong>in</strong> <strong>the</strong> conta<strong>in</strong>ed greenhouse (KKS/KBS/KML) 2007<br />

Development <strong>of</strong> pigeonpea transgenic events for enhanced level <strong>of</strong> ß-carotene: Agrobacterium-mediated genetic<br />

transformation is be<strong>in</strong>g carried out regularly us<strong>in</strong>g <strong>the</strong> b<strong>in</strong>ary vectors conta<strong>in</strong><strong>in</strong>g maize psy1 gene driven by oleos<strong>in</strong><br />

promoter for generat<strong>in</strong>g pigeonpea transgenic events with enhanced level <strong>of</strong> ß-carotene. <strong>The</strong> putative transgenic<br />

pigeonpea shoots obta<strong>in</strong>ed under antibiotic selection pressure are be<strong>in</strong>g elongated for root<strong>in</strong>g. About 40 putative<br />

transgenic plants with maize psy1 have been transferred to <strong>the</strong> conta<strong>in</strong>ment greenhouse for fur<strong>the</strong>r analysis. <strong>The</strong><br />

primers specific to <strong>the</strong> cod<strong>in</strong>g sequence <strong>of</strong> psy1 genes were designed and conditions for PCR amplification were<br />

optimized. <strong>The</strong> primary T 0 putative pigeonpea plants are be<strong>in</strong>g molecularly analyzed us<strong>in</strong>g PCR for <strong>the</strong> presence <strong>of</strong><br />

psy1 gene.<br />

Development <strong>of</strong> pigeonpea transgenic events for enhanced level <strong>of</strong> methion<strong>in</strong>e: In separate studies, b<strong>in</strong>ary gene<br />
