Premessa: There is increasing interest about HIV-1 RNA quantification in samples from<br />

virologically suppressed patients with HIV-1 RNA below 40cp/ml. It has been described<br />

that, at the lower limit of quantification, concordance between commercial assays is weak.<br />

Obiettivo: We compared the performance of Abbott Real-time HIV-1 assay and Roche<br />

COBAS/TaqMan HIV-1 v2.0 for quantification of HIV-1 RNA in clinical samples with viral<br />

load values ranging from 40 to 500cp/ml. Moreover, the concordance on the ability to<br />

detect the presence of HIV-1 RNA in samples resulted below 40cp/ml with the Abbott<br />

assay was also analysed.Quantitative results were compared using correlation, linear<br />

regression and Bland&Altman analyses of log10-transformed HIV-1 RNA copy numbers.<br />

Concordance on the ability to detect HIV RNA was measured by Cohen‟s kappa statistics.<br />

Risultati: A good correlation between assays was observed in samples with viral load<br />

values ranging from 40 to 500 cp/ml (Pearson r=0.8540; p=0.0001): Abbott and Roche<br />

assays measured a mean value of viremia of 2.003 + 0.376 log10cp/ml and 2.038 + 0.240<br />

log10cp/ml, respectively, and quantitative differences did not exceed 0.3 log10cp/ml.<br />

Below 40cp/ml, the degree of concordance between the two assays for samples given as<br />

“detected
