262 SEC Option<br />

Amount Values<br />

In this section, you can enter values that affect how the concentrations<br />

are calculated. If you are making a single data acquisition prior to<br />

calibrating your method, simply leave these values at the default level.<br />

Sample Amount<br />

The Sample Amount value is used as a divisor during calculation of<br />

concentrations. It is intended to compensate for differences between<br />

samples due to weighing and when percentages of the total sample are<br />

being calculated rather than the amount detected in an injection.<br />

Internal Standard Amount<br />

For calibration runs, the Internal Standard Amount is taken from the<br />

method Peak Table. For unknown runs, enter the amount of the Internal<br />

Standard in your unknown sample.<br />

Multiplication Factor<br />

Enter a multiplication factor to be used for this run. All analyzed peaks<br />

will be multiplied by this factor.<br />

Calibrate<br />

This box is only used for calibration of partition chromatography samples.<br />

It is not used in SEC runs.<br />

When you have completed the Single Acquisition Run dialog box, click<br />

Start to begin the acquisition. The current data will appear in the<br />

chromatogram window as it is acquired and stored on disk. At the end of<br />

the run, the chromatogram will be analyzed according to the method<br />

parameters, and a report generated if specified. If the sample is not<br />

analyzed at the end of the acquisition, click the Analyze button if you<br />

wish to view the results.<br />

Sample Description<br />

When you click the Description button, you can enter a text description<br />

for the sample that is saved in the data file. The description can be<br />

viewed from the Open File dialog, or when the file is open as the current<br />

data file using the Data/Properties command.<br />

Submit<br />

This button appears when data is currently being acquired using a<br />

sequence or single run. The Submit button allows you to submit a single<br />

acquisition to be run at the completion of the current run. The run is<br />

entered at the end of the Run Queue if you are currently running a<br />

sequence of runs.<br />

Submit Priority<br />

This button will appear when data is currently being acquired using a<br />

sequence. When you click Submit Priority… you can submit the single<br />

run to be executed immediately after the current sequence run record in<br />

the Run Queue. After this sample is finished, the sequence will resume.<br />

If the chromatogram is not integrated at the end of the run, or if you<br />

were expecting a report and none was printed, check the Method/<br />

Properties section of your method to make sure data analysis and<br />

reporting are turned on for this method.
