General Results 1993-1995:<br />

Cell hybrids between embrionic stem cells and mouse Peucocytes were<br />

obtained during differentiation for study <strong>of</strong> organization and<br />

inactivation mechanism <strong>of</strong> mammilian X-chromosome. Using <strong>the</strong>m<br />

<strong>the</strong> ability <strong>of</strong> a differentiated cell <strong>to</strong> dedifferentiate till <strong>the</strong><br />

pluripotential state while bringing it in<strong>to</strong> an embryonal stem cell was<br />

demonstrated for <strong>the</strong> first time. It is shown that a short-term rize <strong>of</strong><br />

DNA one-chain breaks and sister chromatid exchanges can be seen in<br />

2-3 miotic divisions after <strong>the</strong> beginning <strong>of</strong> spontaneous<br />

differentiation <strong>of</strong> embryonal stem cell in vitro as well as at<br />

differentiation introduction in tera<strong>to</strong>carcynoma cells.<br />

At normal preimplantational mouse development <strong>the</strong> level <strong>of</strong> DNA<br />

breaks is rizing at <strong>the</strong> 8-16 blas<strong>to</strong>meres stage. Works on obtaining<br />

transgenic animals in a carnivora representative American mink are<br />

carried out.<br />

Using cell hybrids, by methods <strong>of</strong> electrophoretic division <strong>of</strong> proteins<br />

and Sau<strong>the</strong>rn blot hybridization about 80 genes on an American mink<br />

chromosome, about 20 genes on a common shrew chromosome and 4<br />

genes on a domestic pig chromosome are assigned.<br />

A collection <strong>of</strong> cell lines, containing one chromosome <strong>of</strong> pig 2, 5, 8, 12<br />

or t(1, 13) against <strong>the</strong> background <strong>of</strong> mink chromosomes was<br />

obtained as well as a collection <strong>of</strong> human monochromosomal hybrid<br />

lines with chromosomes 3, 5, 12, 13, 15, 18, 21 and X. These lines<br />

are a valuable source <strong>of</strong> precise physical mapping <strong>of</strong> mammal<br />

chromosomes and obtaining chromosome-specific libraries.<br />
