México, D.F. 04510 México.RESUMENEl objetivo de este estudio fue desarrollar las modificaciones necesarias para mejorar las prestaciones delELISA indirecto de paratuberculosis para la especie ovina. En particular, se evaluó la sustitución del conjugadoespecífico de especie por Proteina G, con el fin de disminuir de esta forma el ruido de fondo y aumentarla sensibilidad. Para la puesta a punto de la técnica se utilizaron tres controles positivos de diferentegrado de reactividad, los cuales fueron diluidos de forma seriada y enfrentados a diferentes concentracionesde conjugado, de manera que se pudiese definir la concentración óptima tanto de suero como de Proteina G.Una vez optimizado el protocolo laboratorial que proporcionaba la mejor relación respuesta/ruido se validóla eficacia diagnóstica del ELISA en una serie de 118 casos clínicos sobre los que se disponía resultadoshistopatológicos y bacteriológicos. Así, considerando como referencia solamente los animales con resultadosde cultivo de tejidos se estimó una sensibilidad del 100% y una especificidad del 77,0%. Teniendo encuenta solamente los resultados histopatológicos, las estimaciones de sensibilidad y especificidad fueron del79,2% y 89,1%, respectivamente. La combinación de ambas técnicas de referencia mostró los mejores resultadosde eficacia diagnóstica del ELISA ya que arrojó unos valores de sensibilidad y especificidad del 96,2%y del 100%, respectivamente.Palabras clave: cultivo, lesión histopatológica, sensibilidad, especificidad.INTRODUCCIÓNEl diagnóstico de la Paratuberculosis, tanto en laespecie ovina como en todas las que se ven afectadaspor este proceso, presenta una serie de dificultadesque derivan en unos casos de la falta deespecificidad, en otros de la escasa sensibilidad, yen otros del elevado costo y tiempo necesario parasu realización.En el caso de la especie ovina, aunque se disponíaya de un método ELISA de eficacia razonablementebuena, se venía observando una reactividad un tantoexcesiva que se atribuía a cierto ruido de fondo inespecífico.Tomando como base el ELISA desarrolladopor nuestros grupos en proyectos anteriores, se probaronuna serie de modificaciones principalmentebasadas en el uso de distintos tipos de conjugados yse procedió a validar la eficacia diagnóstica en suerosde animales de estado paratuberculoso definido porel cultivo y el examen histopatológico.MATERIAL Y MÉTODOSPara la realización del ELISA se emplearon placasmicrotiter de poliestireno de alta capacidad de fijación.Estas placas eran fijadas con antígeno PPA3,siguiendo el método descrito por Aduriz (1993).Para evitar en medida de lo posible las reaccionesinespecíficas se adsorbían los anticuerpos inespecíficosdiluyendo al 50% el suero problema con unasuspensión de M. phlei (Yokomizo et al., 1983;Milner et al., 1987) en solución salina mediante
