(GRAN BRETAÑA).RESUMENEste estudio pretende evaluar la técnica LTR PCR en células de sangre y leche comparándola con la técnicaserológica de elección en la actualidad como es la Inmunodifusión en Gel de Agar (IDGA). Para la realizaciónde este trabajo se recogieron muestras de leucocitos de sangre periférica (LSP) y células de leche (CL)de 109 y 64 ovejas respectivamente previo a la necropsia. La clasificación de los animales se llevó a cabo enbase a su estado serológico (IDGA) en el momento del sacrificio, resultando así tres grupos: Grupo 1: AnimalesIDGA negativos de rebaños libres de la infección (18 ovejas), Grupo 2: Animales IDGA negativos derebaños infectados por el virus MV (27 ovejas) y Grupo 3: Animales IDGA positivos de rebaños infectados(64 ovejas). En el primer grupo ninguno de los animales resultó ser positivo a PCR en LSP (0/18) ni en CL(0/12). En el Grupo 2, 20/27 fueron positivos en LSP y 10/16 en CL. En el Grupo 3, 53/64 animales fueronpositivos en células de sangre y 19/36 dieron resultados positivos en células de leche. Tres de las siete ovejasPCR negativas en sangre en el Grupo 2 fueron PCR positivas en al menos uno de los órganos diana analizados,mientras que 9/11 de las ovejas PCR negativas en sangre del Grupos 3, presentaron resultados de PCRpositivos en alguna de las muestras de tejido procesadas.Palabras clave: lentivirus, diagnóstico, Reacción en Cadena de la Polimerasa.INTRODUCCIÓNEl Maedi-Visna es una enfermedad de declaraciónobligatoria que figura en la lista B de la OIE(Oficina Internacional de Epizootias) y que precisade normas concretas para la comercialización y libreintercambio de animales con el resto de países europeos(OIE, Manual 1996). El Maedi es una enfermedadendémica en la cabaña ovina de la CAV ymuy pocos rebaños se pueden considerar libres deella (González, 1989). Las pérdidas económicas quegenera son variadas y difíciles de cuantificar debidoa la aparición lenta y progresiva de la enfermedad:bajas por casos clínicos, aumento de la tasa de reposición(Houwers et al, 1990), disminución en la producciónláctea (Snowder et al, 1990), disminuciónen la ganancia de peso vivo del cordero (Kirton etal, 1976), costes de tratamientos sintomáticos, etc.En la actualidad la técnica de elección en el diagnósticodel Maedi-Visna es la Inmunodifusión enGel de Agar, pero esta técnica presenta una serie dedesventajas como son la dificultad de interpretación(requiere personal especializado), el uso de un antígenosemipurificado (el cual es caro de producir ydifícil de estandarizar) (Kwang et al., 1993) y porotro lado es una técnica de sensibilidad relativa a lahora de detectar la infección en los primeros estadioso bien en animales con bajo nivel de anticuerpos.Las posibilidades que ofrece la biologíamolecular en el diagnóstico de los lentivirus ensangre (Wagter, L. et al., 1997) y leche (Vitu et al.,1997) han permitido desarrollar una técnica de PCRque es capaz de detectar el ADN proviral del VMVinsertado en el genoma de la célula hospedadora(células del sistema monocito-macrófago), factor
