as 5HTT as well as other encoding components of the 5HT system,
such as TPH2, are also linked to Neuroticism and Cluster C personality
disorders, suggesting a developmental trajectory from increased
Neuroticism over anxious-fearful personality disorders eventually
leading to clinically relevant major depression. This development
might well be shaped and accelerated by adverse life events To date
however, there are almost no studies which specifically test this hypothesis,
as pertinent longitudinal studies are a daunting task which
nevertheless will be accomplished in the near future.

Introduction: Depressive disorders represent a major socioeconomic
and therapeutic challenge and are associated with a high degree
of individual burden and distress. The heritability of depression has
been estimated to be 37 %. Previous studies have implicated dysregulations
of the HPA-axis in the pathogenesis of depressive disorders.
Recently, an interaction between childhood abuse and polymorphisms
within the Corticotropin-Releasing Hormone Receptor
Gene (CRHR1) was reported, connecting early life stress and genetic
susceptibility to adult depression. We tested the hypothesis of an
interaction of childhood maltreatment and neglect with polymorphisms
and haplotypes within the CRHR1 gene in a general population
sample.
Method: All participants (n=1638) were Caucasian subjects from
Study of Health in Pomerania (SHIP). The gene by environment
interactions between 34 single nucleotide polymorphisms (SNP) at
the CRHR1 locus and measures of child abuse and neglect (Childhood
Trauma Questionnaire) on adult depressive symptoms were
investigated.
