E<strong>in</strong>leitung: Das Antikonvulsivum Valproat ist e<strong>in</strong>e wichtige pharmakologische<br />
Option <strong>in</strong> <strong>der</strong> Behandlung bipolarer Störungen und<br />
dabei <strong>in</strong>sbeson<strong>der</strong>e <strong>der</strong> Verlaufsform des „Rapid Cycl<strong>in</strong>g“ (RC). Es<br />
herrscht jedoch nach wie vor Unklarheit über den Wirkungsmechanismus<br />
dieses Medikaments.<br />
Methode: In dieser Studie wurde die Erregbarkeit des motorischen<br />
Kortex mittels Transkranieller Magnetischer Stimulation (TMS)<br />
und <strong>der</strong> Ableitung motorisch evozierter Potentiale (MEPs) vor und<br />
nach Gabe von 800 mg Valproat bei 15 gesunden Probanden gemessen.<br />
Es wurden dazu die TMS Parameter „Rest<strong>in</strong>g Treshold“<br />
(RT), „Cortical Silent Period“ (CSP), „Short Intracortical Inhibition“<br />
(SICI), „Intracortical Facilitation“ (ICF) und „MEP Stimulus<br />
Response“ (MEP-SR) bestimmt.<br />
Diskussion / Ergebnisse: Nach Gabe von Valproat zeigt sich für die<br />
MEP S-R e<strong>in</strong>e signifikante Verän<strong>der</strong>ung, die übrigen untersuchten<br />
Parameter bleiben unverän<strong>der</strong>t.. Überraschen<strong>der</strong>weise erhöht Valproat<br />
die mittlere MEP Antwort auf TMS Stimuli verschiedener<br />
Intensität signifikant (p= 0.01). Valproat unterscheidet sich damit<br />
<strong>in</strong> se<strong>in</strong>em E<strong>in</strong>fluss auf die Erregbarkeit des Kortex von allen an<strong>der</strong>en<br />
bekannten Antiepileptika, Neuroleptika und Benzodiazep<strong>in</strong>en.<br />
Die Erhöhung <strong>der</strong> MEP deutet darauf h<strong>in</strong>, dass Valproat nach e<strong>in</strong>maliger<br />
Gabe die synaptische Übertragung bee<strong>in</strong>flusst. Ähnliche<br />
Effekte auf die kortikale Erregbarkeit f<strong>in</strong>den sich nach Gabe dopam<strong>in</strong>erger<br />
sowie noradrenerger Substanzen. Inwieweit die beobachtete<br />
Wirkung von Valproat mit <strong>der</strong> antikonvulsiven und stimmungsstabilisierenden<br />
Wirkung im Zusammenhang steht, muss <strong>in</strong><br />
weiteren Studien geklärt werden.<br />

It is well known that menthol evokes a sensation of cold when applied<br />
to sk<strong>in</strong> or mucosa. From tobacco <strong>in</strong>dustry it is known as an<br />
additive substance <strong>in</strong> cigarettes enhanc<strong>in</strong>g the potential of addiction.<br />
The mechanism by which menthol activates cold receptor<br />
neurons or even modulates higher cognitive functions is still un<strong>der</strong><br />
debate. Beside activation of TRPM8 channels, blockade of Ca2+<br />
currents with reduction of Ca2+ dependent K+ currents have been<br />
discussed. Our whole cell patch-clamp record<strong>in</strong>gs on the GnRH<br />
produc<strong>in</strong>g hypothalamic mouse cell l<strong>in</strong>e GT1-7 demonstrates a<br />
pronounced reduction of an outward K+ current by menthol, which<br />
can not be expla<strong>in</strong>ed by the conventional assumptions about the<br />
action of menthol via TRP channels. Our data po<strong>in</strong>t to a dose dependent<br />
completely reversible <strong>in</strong>hibition of voltage-dependent slow<br />
delayed rectifier K+ channels (IK(DR)). Furthermore the menthol<br />
effect on outward currents is largely <strong>in</strong>dependent of extracellular<br />
Ca2+, which rules out an exclusive <strong>in</strong>volvement of TRP and Ca2+<br />
activated K+ channels. At higher menthol concentrations an additional<br />
block of the IK(A) currents is obta<strong>in</strong>ed, while Na+-currents<br />
rema<strong>in</strong> unaffected. As menthol blocks the DR-type K+ current, it<br />
changes the shape, fir<strong>in</strong>g pattern and function of action potentials.<br />

Furthermore it may lead to a AP change from neuronal-like <strong>in</strong>formative<br />
click to a more endocr<strong>in</strong>ological-like or<strong>der</strong> to secrete vesicles<br />
of hormones. While the delayed rectifiers are housekeep<strong>in</strong>g<br />
channels and expressed <strong>in</strong> more or less every neuron <strong>in</strong> different<br />
amounts, next to cold-sens<strong>in</strong>g, menthol has a second function: it<br />
plays a tun<strong>in</strong>g role <strong>in</strong> neuroendoc<strong>in</strong>ological systems with K+ channels<br />
as specific targets.<br />

Introduction: A formulation of a cytok<strong>in</strong>e model of cognitive<br />
function un<strong>der</strong> immunologically unchallenged physiological conditions.<br />
Method: The proposed cytok<strong>in</strong>e model of cognitive function un<strong>der</strong><br />
unchallenged conditions is based on empirical work by this group<br />
<strong>in</strong>vestigat<strong>in</strong>g the effects of cytok<strong>in</strong>es at the prote<strong>in</strong> and genetic level<br />
on cognitive function as well as experimental animal models <strong>in</strong>vestigat<strong>in</strong>g<br />
the effects of TNF <strong>in</strong> transgenic mice.<br />
Discussion / Results: In a study among 369 healthy el<strong>der</strong>ly from<br />
the general population, we found the chemok<strong>in</strong>e IL-8 to be significantly<br />
associated with poor cognitive performance <strong>in</strong> the memory,<br />
attention and motor doma<strong>in</strong>s. In a similar study among 369 healthy<br />
<strong>in</strong>dividuals, genetic variants of IL-1beta were related to poor memory,<br />
whereas TNF-alpha was related to better cognitive speed performance.<br />
In contrast, genetic variants of IL-6 showed no association<br />
with cognitive performance <strong>in</strong> humans. Animal studies of our<br />
group show that the absence of TNF (B.6TNF-/-; N=10) is detrimental<br />
of cognitive function dur<strong>in</strong>g neurodevelopment, while dur<strong>in</strong>g<br />
ag<strong>in</strong>g the absence of TNF (B.6TNF-/-; N=10) is related to improved<br />
cognitive performance as opposed to wt mice (C57BL/6;<br />
N=10). The cytok<strong>in</strong>es IL-1β, IL-6 and TNF-α have effects on complex<br />
cognitive processes at the molecular level, such as synaptic<br />
plasticity, neurogenesis, as well as neuromodulation. The f<strong>in</strong>d<strong>in</strong>gs<br />
provide evidence for a cytok<strong>in</strong>e model of cognitive function, which<br />
shows that cytok<strong>in</strong>es play an <strong>in</strong>timate role <strong>in</strong> the molecular and cellular<br />
mechanisms sub-serv<strong>in</strong>g learn<strong>in</strong>g, memory and cognition un<strong>der</strong><br />
physiological conditions.<br />

den Zusammenhang zwischen Alkoholkonsum und erhöhten<br />
Homocyste<strong>in</strong>spiegeln zeigen. Diese Am<strong>in</strong>osäure bee<strong>in</strong>flusst den<br />
humanen Methylierungsstoffwechsel. So f<strong>in</strong>det man bei Alkoholabhängigen<br />
verän<strong>der</strong>te Methylierungsmuster auf genomweiter wie<br />
auch genspezifischer Ebene. Ziel <strong>der</strong> vorliegenden Studie bestand<br />
<strong>in</strong> <strong>der</strong> Analyse genetischer Polymorphismen von am C1-Stoffwechsel<br />
beteiligten Enzymen und <strong>der</strong>en E<strong>in</strong>fluss auf Plasma Homocyste<strong>in</strong><br />
(PH) sowie auf genomweite DNA Methylierung (gDNA-M)<br />
bei Alkoholabhängigkeit.<br />
Methode: Es wurden 106 Patienten (86 Männer, 20 Frauen), die zur<br />
Alkoholentzugsbehandlung stationär aufgenommen wurden, <strong>in</strong> die<br />
Studie e<strong>in</strong>geschlossen. Alle erfüllten die Kriterien e<strong>in</strong>er Alkoholabhängigkeit<br />
nach DSM-IV. Die statistische Analyse erfolgte mittels<br />
e<strong>in</strong>es multivariaten l<strong>in</strong>earen Modells. Auf genetischer Ebene analysierten<br />
