EFFECT OF CHITOSAN-COATED GOLD NANOPARTICLES (AUNPS)<br />

Nanotechnology employment on devices and medical treatments is a growing<br />

study trend since the past years. Due to its biocompatibility and low toxicity, gold<br />

nanoparticles (AuNps) represent an attractive therapeutic alternative against<br />

cancer, anti-virus, immune-stimulating and for tissue regeneration,<br />

nevertheless, just as conventional drugs, studies for cytotoxicity, side effects,<br />

and immunological response are required. Dendritic cells (DC) represents an<br />

excellent study model to evaluate the immune system reaction to an external<br />

agent; these cells are considered as the bridge between the innate and adaptive<br />

immunity since they can trigger and specific defense through Toll-like receptors<br />

(TLR). The aim of this work is to determinate the effect of AuNps on DC, for this<br />

AuNps were synthesized using the Turkevich method, employing chitosan as<br />

reducing agent. The size was analyzed by dynamic light scattering (DLS) and the<br />

surface plasmon resonance, was determined by UV-vis spectroscopy.<br />

Peripherical blood mononuclear cells (PBMCs) viability was evaluated by flow<br />

cytometry, as well as phagocytic capacity and maturation markers expression<br />

(CD80, CD83, HLA and, CD14). Average size was found in 22nm. The plasmon<br />

surface resonance was observed at 520 nm maximums absorbance. 10, 20, 30<br />

y 40 µM AuNps concentrations were evaluated, and cytotoxicity was not<br />

observed at concentrations below 30 µM, same results were shown in response<br />

to chitosan and chloroauric acid, synthesis precursors.<br />
