TELODENDRIMERS BASED NANOSTRUCTURES FOR DRUG<br />

DELIVERY: A HYBRID PLATFORM COMBINING ADVANTAGEOUS<br />

TRAITS OF LINEAR POLYMERS WITH DENDRIMERS<br />

Macromolecules based therapeutics (nanomedicine) continues to evolve linear<br />

block-copolymers to branched (miktoarm polymers) and hyperbranched<br />

(dendrimers) architectures. It is becoming increasing clear that combining<br />

important characteristics of linear polymers with those of branched and<br />

hyperbranched architectures could yield hybrid structures (telodendrimers) that<br />

could help address key issues in nanomedicine. Despite tremendous potential,<br />

significant challenges related to simplifying their synthetic schemes,<br />

understanding their self-assembly behavior, and enabling them to multi-task,<br />

still remain. An important parameter for constructing these scaffolds is to<br />

develop synthetic tools that can provide desired spatial distribution of different<br />

components in these nanostructures. The methodologies need to be simple and<br />

highly versatile, and adaptable to the needs of biology. We shall describe how<br />

chemical innovation can provide easy access to a variety of telodendrimers, and<br />

their self-assembly could be tuned with varied compositions. Such functional<br />

nanostructures can be used for monotherapy through physical encapsulation<br />

or drug conjugation, or combination therapy using multiple drugs. Their<br />

structural complexity is ideally suited to adapt to the needs of ever-increasing<br />

complexity of high morbidity rate diseases and facilitate drug delivery at specific<br />

disease sites.<br />
