PREPARATION AND IN VITRO EVALUATION OF A<br />

RADIOLABELLED NANOCARRIER AS AN APPROACH FOR<br />

COMBINED THERAPY ON TRIPLE NEGATIVE BREAST CANCER<br />

CELLS<br />

Introduction: Nanoradiopharmaceuticals have acquired more attention as<br />

suitable approaches for theranostic applications in several cancer tumors.<br />

Polymeric nanoparticles have generated increasing interest in this field due to<br />

their ability to encapsulate drugs and their ability for targeted a controlled drug<br />

delivery. Paclitaxel (PTX) has suggested remain as the standard option to treat<br />

triple negative breast cancer (TNBC). Several side effects have been reported on<br />

this treatment. Bombesin has demonstrated high sensitivity to detect GRPr<br />

(Gastrin Release Peptide receptor) in all phenotypes of breast cancer. Triple<br />

Negative Breast Cancer (TNBC) represents the worst prognosis.<br />

Objective: The aim of this study was to obtain the 177 Lu-Bombesin-PLGA(PTX)<br />

nanosystem as an approach potentially useful for combined therapy on triple<br />

negative breast cancer cells through paclitaxel (PTX) targeted delivery in MDA-<br />

MB-237 cells which overexpress GRPr and the radiotherapy produced by the β -<br />

energy of Lutetium-177 on target site.<br />

Methods: Nanoparticles were synthetized and characterized by SEM, FT-IR and<br />

DLS. The Bombesin-PLGA(PTX) was radiolabelled and the cytotoxicity was<br />

evaluated on MDA-MB-237 TNBC.<br />

Results: The Bombesin-PLGA(PTX) obtained nanoparticles showed a spherical<br />
