PLLGA COPOLYMERS: A DEGRADATION, DRUG DESORPTION AND<br />

CYTOTOXICITY STUDY<br />

The aims of the present work, were to study the poli(L-lactic-co glycolic)<br />

copolymers (PLLGA) degradation process, prednisone drug desorption and<br />

PLLGA degradation products cytotoxicity. For these reason three combinations<br />

of (PLLGA) with different lactide to glycolide ratio (70:30, 80:20 and 90:10 %wtL-<br />

LA:%wtGA ) were synthesized by means of ring opening polymerization. The<br />

materials obtained were characterized in order to know their structure, chemical<br />

composition, molecular weight and glass transition temperature. The<br />

degradation and drug desorption studies were realized in vitro during 336h at<br />

37°C in saline solution. For degradation study change in molecular weight, mass<br />

loss and optical microscopy images were taken into account, and for prednisone<br />

desorption UV spectroscopy at 240nm was used. The results showed that PLLGA<br />

degradation followed a first order kinetic and prednisone released was<br />

government by two phenomena diffusion and degradation. For these reason<br />

the drug desorption kinetic was adjusted to two model proposed for<br />

biodegradable systems: Higuchis and Charlie. Drug diffusion coefficients were<br />

higher in copolymers with higher percentage of glycolic acid, and they were<br />

increasing, as the degradation process was progressing. The cytotoxicity assay<br />

evidenced that PLLGA copolymers and PLLGA degradation products were not<br />

toxic in endothelial cells.<br />
