Bacteria Isolation from Water Sample of Seahorse Cultivation Tank Before
and After Filtration

A study on water microbiology quality was conducted at seahorse nursery of Aquatrop
Hatchery. Samples from the water supply before the filtration system at the reservoir
and after the filtration system prior to the displacement into the tanks. Isolation was
done using sterile transport swab which was then inoculated onto the MNA and EMB
agar using the serial dilution and MPN method. Pure culture was made to identify the
bacteria. Results showed that formation of metallic colonies on EMB agar from the
sample of water before the filtration system. Pathogenic bacteria Escherichia coli was
identified. The filtration system is successful in filtering pathogenic bacteria, but
improvements can be made for a better microbial water quality.
